Hexobarbital-Metabolizing Enzyme Induction Study in Mice

Fifty male and 50 female mice were assigned randomly to one of five treatment groups (10
mice of each sex/treatment group). In each sex, three groups of mice orally received one of three
graded dosage levels of an investigational drug (10, 50, 250 mg/kg), one group orally received
0.9% sterile Saline (25 ml/kg) as a negative control, and one group received Sodium Phenobarbital
(75 mg/kg) injected intraperitoneally. Sodium Phenobarbital was included in the experiment as a
positive control since it is known to include hexobarbital metabolizing enzymes. Treatment was
administered b.i.d., approximately four-and-one half hours apart, over a period of four consecutive
days. A hexobarbital (100 mg/kg) injection was given intraperitoneally approximately 24 hours
following the last dose of the assigned treatment. Each animal's sleeping time following
hexobarbital administration was measured as the duration of time (minutes) from the loss of the
righting reflex to the time of spontaneous righting. One male mouse (50 mg/kg) died from an
intubation accident on the fourth day of treatment. If drug administration induces hexobarbital-metabolizing enzymes, then the length of sleep should decrease with increase in dose of drug.
